摘要 目的观察人大肠癌和正常大肠黏膜组织中T淋巴瘤侵袭转移诱导因子1(T lymphoma invasion and metastasis inducing factor1,Tiam1)和RacGTP酶激活蛋白1(Rac GT-Pase activating protein1,Rac1)的表达情况并探讨其临床意义。方法应用SP免疫组化法检测50例经4%中性甲醛固定和石蜡包埋的大肠癌及50例正常大肠黏膜组织标本中Tiam1和Rac1蛋白的表达情况并分析其与大肠癌临床病理参数间的关系。结果Tiam1和Rac1蛋白在正常大肠黏膜组织中均呈阴性染色(0/50,0),但在大肠癌组织中则呈阳性染色(42/50,84.00%),两者间差异有统计学意义,P=0.003。Tiam1和Rac1蛋白染色阳性率在高分化(9/13,69.23%)和低分化(14/14,100.00%)之间、T1+T2或Ⅰ+Ⅱ期(8/13,61.54%)和T3+T4或Ⅲ+Ⅳ期(34/37,91.89%)之间、无淋巴结转移(15/22,68.18%)和有淋巴结转移(27/28,96.43%)之间的差异均有统计学意义(P值分别为0.025,0.01,0.007),但Tiam1和Rac1蛋白表达水平与大肠癌患者的性别、年龄、癌变原发部位及癌肿大小无关(P值分别为0.686,0.975,0.791,0.709),差异无统计学意义。结论Tiam1和Rac1蛋白的表达与人大肠癌浸润和转移密切相关。 OBJECTIVE: To investigate the expressions and clinical significance of T lymphoma invasion and metastasis inducing factor 1 (Tiam1) and Rac GTPase activating protein 1 (Rac1) in human colorectal cancer (CRC) and normal colorectal mucosa. METHODS: The expressions of Tiaml and Racl proteins in formalin-fixed, paraffin-embedded specimens of colorectal cancer and normal colorectal mucosa of 50 cases respectively were detected by using SP immunohistochemical method. Simultaneously its relationship to the clinicopathological parameters of colorectal cancer was analysed. RESULTS: There was negative staining of Tiaml and Racl proteins in normal colorectal mucosa (0/50, 0), while positive staining was detected in colorectal cancer (42/50, 84.00% ). There were significant differences between them(P= 0. 003). Accompanyed with differentiated degree of histologic type descending, depth of invasion increasing, stage of TNM upgrading and lymph node metastasis happening, the positive staining rates of Tiam1 and Rac1 proteins in colorectal cancer significantly ascended (P=0. 025,0.01,0.01,0. 007,respectively), but there was no obvious correlation between its expression and the sex, age of patient, primary pathological location or size of colorectal cancer (P=0.686, 0.975, 0.791, 0.709, respectively). CONCLUSION: The expressions of Tiaml and Racl proteins in human colorectal cancer are obviously correlated with the clinical stage, the degree of differentiation and tumor metastasis.
