摘要
将来源于菊芋的蔗果寡糖基因作为目的基因,抗卡那霉素的NTPⅡ基因作为选择标记基因,利用下胚轴和子叶做外植体与农杆菌共同培养,在高代自交的番茄中,获得卡那霉素的抗性植株。PCR检测结果表明,电泳图有蔗果寡糖基因的扩增带,抗性株含蔗果寡糖基因,证实所得的卡那霉素抗性株为转蔗果寡糖基因番茄。
Based on Fructooligosaccharide gene and a selectable marker gene NTP II which confers resistance to the Kanamycin, Tomato Short Chain Fructooligosaccharide are acquired via tissue culture in vitro using the cotyledonary and hypocotyl segments as the explans. Kanamycin transformants are obtained in Lycopersicon esculentum varieties. PCR results show that there are Fructooligosaccharide extending strips in the electrophoresis map and the tomato plants are positive with perfect resistance to the Kanamycin with Fructooligosacchadde.
