用同一皮肤标本培养、纯化人正常黑素细胞、角质形成细胞及成纤维细胞的观察

摘要 目的建立从同一皮肤标本同步培养人正常黑素细胞、角质形成细胞和成纤维细胞的技术。方法利用同一供体的包皮,表真皮分离后,置37℃,5%CO2的孵箱中培养,根据细胞生长状况进行相应的传代和纯化处理。结果获得大量纯化、生长良好的人正常黑素细胞、角质形成细胞和成纤维细胞。结论实现了对同一组织来源的三种细胞在不同培养基中生长状况的同步观察。 Objective To establish the technique of Culture of normal human melanocyte,keratinocyte and fibroblast isolated from the same donor. Methods The epidermis was detached from foreskin by trypsinization. The pellets were resuspended into melanocyte conditional medium, keratinocyte conditional medium and fibroblast medium, accordingly. After counting, the cells were seeded onto plate at 1.0 × 10^4 -2.0 × 10^4/cm^2 and incubated at 3722,5% CO2 ,respectively. Cells were subcuhured according to growth. Results We obtained the cultures of melanocyte, keratinocyte and fibroblast. Conclusion we carried out the observation of biology of three cells under different culture conditions.
