白芍总苷对IL-1α诱导胶原性关节炎大鼠成纤维样滑膜细胞功能的影响 被引量:25

Effects of TGP on functions of fibroblast-like synoviocytes under the stimulation of IL-α in CIA rats

摘要 目的研究白芍总苷(TGP)对rIL-1α诱导胶原性关节炎大鼠成纤维样滑膜细胞(FLS)的影响,并探讨其部分机制。方法鸡Ⅱ型胶原(CCⅡ)诱导大鼠胶原性关节炎(CIA);MTT法检测FLS的增殖反应;放射免疫法(RIA)测定FLS产生PGE2和cAMP的水平。结果在rIL-1α(0.01、0.1、1、10μg.L-1)和1μg.L-1rIL-1α(3、6、12、24h)体外刺激下,CIA大鼠FLS产生的PGE2和cAMP水平均升高;TGP(2.5、12.5、62.5、125、250mg.L-1)体外给药可不同程度地抑制FLS的增殖反应和PGE2产生,而进一步升高cAMP水平。结论在不同浓度同一时间和同一浓度不同时间的rIL-1α体外刺激下,FLS产生PGE2和cAMP水平均升高;TGP体外作用FLS可抑制其增殖和产生PGE2,进一步升高cAMP的水平。 Aim To study effects of TGP on fibroblast- like synoviocytes(FLS) under the stimulation of rIL-1α and investigate the mechanism. Methods CIA was induced in male Sprague-Dawley rats immunized with chicken type Ⅱ collagen in Freund's complete adjuvant; rIL-1α-induced FLS proliferation was observed by 3-(4,5-dimethylthiazol-2-yl ) 2,5-diphenyhetrazolium bromide(MTT) assay. Levels of PGE2 and cAMP were determined by radioimmunoassay(RIA). Results Levels of PGE2 and cAMP produced by FLS under the stimulation of rIL-1α(0. 01,0. 1,1,10 μg· L^-1 ) and rIL-1α (3,6,12,24 h ) were elevated. TGP ( 2.5,12. 5,62.5,125,250 mg· L^-1 ) inhibited the proliferation and decreased the level of PGE2 and increased the level of cAMP. Conclusion Under the stimulation of rIL-1α in different time and different concentration, levels of PGE2 and cAMP produced by FLS were elevated significantly. TGP inhibited the proliferation and decreased the level of PGE2 and increased the level of cAMP in vitro.
