大豆苷元磷酰化产物的结构确定 被引量:5

The Phosphorylation Reaction between Daidzein and Di-isopropyl Phosphite Studied by NMR and ESI-MS/MS

摘要 将二烷基亚磷酸酯和四氯化碳混合液滴加到由大豆苷元,N,N-二甲基甲酰胺,三乙胺组成的混合液中,结果表明在该条件下大豆苷元和二烷基亚磷酸酯发生了磷酰化反应,利用核磁共振(NMR)技术,确认单磷酰化大豆苷元为7-二烷基大豆苷元磷酸酯,证明了大豆苷元的7-位羟基在磷酰化反应过程中较4′-位羟基具有更高的反应活性,其目标化合物的结构进一步被电喷雾质谱(ESI-MS/MS),特别是X-ray晶体衍射结果所证实. The Atheron-Todd reaction has been used extensively for synthesis of phosphates and phosphoroamidates. In this study, we showed that daidzein can be phosphorylated by a modified Atheron-Todd procedure in which dialkyl phosphite and tetrachloromethane mixture was dropped into a mixed solution of daidzein, trithylamine and DMF. The reaction product was obtained with good yield, whose structure was determined by NMR, ESI-MS and X-ray crystallography.
