独立生长因子1在内毒素耐受小鼠肺组织中的作用研究

摘要 目的:研究独立生长因子1(Gfi1)在内毒素耐受小鼠接受大剂量内毒素(LPS)刺激后的表达。方法:74只C57BL/6小鼠随机分两组,对照组用0.9%NaCl注射后用大剂量LPS刺激,内毒素耐受组用小剂量LPS预处理后用大剂量LPS刺激。比较两组小鼠存活率,各时间点肺组织病理。用ELISA法检测肺组织TNF-α,RT-PCR检测肺组织Gfi1 mRNA,免疫组化法检测肺组织Gfi1蛋白表达。结果:内毒素耐受组小鼠存活率高,肺组织炎症反应轻,LPS刺激后TNF-α水平未见上升;LPS刺激后2h,6h小鼠肺组织Gfi1的mRNA表达较对照组高(P<0.01);LPS刺激后6h、12h免疫组化显示小鼠肺组织Gfi1表达比对照组高(P<0.01)。结论:内毒素耐受时,大剂量内毒素引起的肺部炎症反应较轻可能与肺部Gfi1高表达有关。 Objective: To study the pulmonary Growth Factor Independence 1 (Gfil)expression in endotoxin tolerance mice. Methods:Seventy-four C57BL/6 mice were randomized into normal control (NC) group and endotoxin tolerance (ET) group. The survival rates were compared and the pathology of lungs were detected. The protein production of TNF-α in lungs was measured by ELISA. The gene expression of Gfil in lungs was measured by semi-quantitative RT-PCR, while the protein production by immuno histochemistry. Results.. In endotoxin tolerance group, the survival rate was higher, the pulmonary inflammatory response was mild and the protein production of TNF-α was not increased. The gene expression of Gfil in lung tissue was higher in endotoxin tolerance group at 2 h, 6 h after high dose LPS challenge (P(0.01). The protein production of Gfil in lung tissue was higher in endotoxin tolerance group at 6 h, 12 h after challenge (P(0.01). Conclusion:The induction of endotoxin tolerance is associated with a profoundly high level of Gfil in lung tissue when challenged with high dose LPS.
