Expression and meaning of platelet activating factor and its receptor in preterm labor

摘要 目的探讨血小板激活因子(PAF)及其受体(PAF-R)在早产和足月产妇母血和胎盘中的表达变化。方法选择2004-05-2005-06在中南大学湘雅二医院及郴州市第一人民医院住院分娩的早产产妇36例,足月产妇30例以及与早产产妇孕周相对应的正常孕妇36例作为对照组。用ELISA法分别测定3组孕产妇母血PAF水平。用免疫组织化学SP法检测早产及足月产组胎盘中PAF-R的表达。结果早产组母血的PAF水平(2.84±0.58)μg/L,高于对照组(2.17±0.48)μg/L及足月产组(2.51±0.57)μg/L,P<0.05。足月产组与对照组比较差异亦有统计学意义(P<0.05)。早产组胎盘组织中PAF-R表达的免疫组化积分(3.12±1.21)高于足月产组(2.47±0.90),P<0.05。早产及足月产组母血PAF水平均与胎盘PAF-R表达呈正相关,早产组中相关系数为r1=0.497,P<0.05,足月产组中相关系数为r2=0.633,P<0.05。结论母血中PAF升高和胎盘PAF-R高表达与早产的发生密切相关,母血中PAF的水平与胎盘中PAF-R的表达呈正相关。 Objective To investigate expression of platelet activating factor (PAF) and its receptor (PAF-R) in blood and placenta of preterm labor. Methods The specimens from 102 women were obrained at the Second Xiangya Hospital of Central South University and the First People Hospital of Chenzhou, between May 2004 and June 2005. They were divided into the following three groups : women ( n = 36 ) in preterm labor, women ( n = 30 ) in term labor, and normal women (n = 36)whose weeks were in equation with preterm labor (control group). ELISA was used to detect the levels of PAF in the pregnant womeng blood ;SP immunohistochemistry was used to detect the expression of the placenta PAF-R in preterm labor and term labor groups. Results The levels of PAF in preterm labor group( 2.84±0. 58) μg/L was higher than that in control group(2.17±0.48)μg/L, P〈0. 05, and also higher than that in term labor group(2.51±0.57) μg/L,P 〈0.05. The statistical significance is found between term labor group and control group (P 〈0. 05 ). The expression of PAF-R in preterm labor group(3.12 ± 1.21 ) was significantly higher than that in term labor group (2.47±0. 90), P 〈0. 05. In the two groups the levels of PAF and the expression of PAF-R were positively correlated with each other. Conclusion The higher levels of PAF in pregnant womeng blood and the higher expression of PAF-R in placenta play an important role in the pathogenesis of preterm labor. There is positive correlation between the PAF levels of pregnant womeng blood and the expression of PAF-R in placenta.
