摘要
目的探讨补阳还五汤及其拆方对脑缺血后髓过氧化物酶(MPO)活性及神经细胞凋亡的影响。方法制备大鼠局灶性脑缺血模型。将SD大鼠随机分为假手术组、模型组、补阳还五汤组、黄芪组、活血组,分光光度法检测各组大鼠脑组织MPO活性,TUNEL法检测各组大鼠脑组织神经细胞凋亡。结果与假手术组相比,模型组缺血侧脑组织MPO活性及TUNEL阳性神经细胞数目显著升高;与模型组相比,补阳还五汤组、黄芪组、活血组缺血侧脑组织MPO活性降低,TUNEL阳性神经细胞数目减少;与拆方组(黄芪组、活血组)比较,补阳还五汤组作用最强。结论补阳还五汤及其拆方可降低脑组织MPO活性,减轻脑缺血后的炎症反应,并可抑制神经细胞凋亡,这可能是补阳还五汤对脑缺血损伤起保护作用的机理之一。补阳还五汤的作用明显优于其拆方,说明方中益气、活血中药配伍具有协同作用。
Objective: To investigate the effects of Buyanghuanwu Decoction (BYHW,补阳还五汤) and the separate components on the activity of MP0 and apoptosis of neuronal cell after cerebral ischemia. Methods: The string inserting method according to Longa' s report was employed to make the model of focal cerebral ischemia. SD rats were divided randomly into the sham -operated group, the model group, the BYHW group, the astragus group and the blood - activating group. The activity of MPO in brain of rats in each the group were analyzed by spec- trophotometry. Neuronal apoptosis of in brain of rats in each group were analyzed by TdT - mediated dUTP - biotin nick end labeling(TUNEL). Results: Compared with those in the sham - operated group, the activity of MPO and the number of positive neuronal cells of TUNEL increased in model group rats' brain tissues; Compared with those in the model group, the activity of MPO and the number of positive neuronal cells of TUNEL in BYHW group, the astragus group and blood - activating group decreased. QDTMT group had the strongest effects compared with astragus group and bolld - activating group. Conclusion: BYHW could lower the activity of MPO in brain tissue, alleviate the inflammary reaction and inhibit neuronal cell apoptosis after cerebral ischemia-reperfusion, which maybe one of mechanisms of protective effects of buyanghuanwu Decoction on cerebral ischemia injury. Effect of BYHW was better than that of separate components groups, which showed that combined use of blood - Activating drug and Qi Reinforcing drug had coordinate effect.
