犬内皮细胞化动脉支架的建立及体外测试

Establishment of a canine endothelialized arterial stent and in vitro test

摘要 目的建立犬血管内皮细胞支架的制作方法,并在体外初步检测其治疗的可行性。方法取犬颈静脉分离培养出血管内皮细胞,并进行体外鉴定。将血管内皮细胞贴附于含有肝素的高分子材料涂层的不锈钢支架表面,形成内皮化支架。采用体外模拟血流冲刷试验观察0—24h冲刷后的细胞形态,并计每1mm长度支架表面的黏附细胞数以检测其黏附能力。结果培养获得犬血管内皮细胞。形成具有完整细胞层的内皮化支架。体外冲刷0、12、24h细胞计数为305.30±22.54、294.70±20.45.288.60±30.97,差异无统计学意义(P〈0.05)。结论犬血管内皮细胞可牢固贴附于含有肝素的药物高分子材料涂层的不锈钢支架表面,形成具有一定抗血流冲刷能力的内皮化细胞支架。 Objective To establish the method of making an endothelialized stem and discuss the feasibility for treatment in vitro. Methods Endothelial cells were isolated and cultured from the canine jugular vein and identified in vitro. Vascular endothelial cells were adhered to the surface of stent coated with heparinized polymer, and an endothelialized stem was established. The stem was washed out with simulated blood at 100 dyne/cm^2 in vitro for 24 h. The morphology of ceils at 0,12,24 h were observed,the cell number on the surface of the stent per mm was counted to test the capability of cell adhesion. Results Canine endothelial cells were obtained. A complete monolayer of endothelial cells was established on the surface of the stent and endothelized stems were made. Cell number counting at 0,12,24 after the beginning of washout was 305.3 ± 22.54,294.7 ± 20.45 and 288.6 ± 30.97 respectively. There was no significant difference. Concision Canine vascular endothelial cells can adhere to the surface of stent coated with heparinized polymer to form an endothelialized stem with the capability of anti-washout.
