摘要
目的:观察四君子汤对利血平所致脾虚大鼠肠道黏膜免疫功能的影响。方法:雄性SD大鼠120只,腹腔注射利血平造成大鼠脾虚模型,四君子汤作为治疗药物,并分高、低剂量组,选用阿托品作为阳性对照药,并设自然恢复组与治疗组相对比。进行小肠PP结计数;并运用流式细胞仪检测PP结中T细胞亚群含量;ABC染色法,并运用图像分析空肠中细胞因子(TNF-α和TGF-β)的表达。结果:各组间在小肠PP结数量及其中的T淋巴细胞亚群无显著差异。模型组大鼠的空肠中TGF-β的表达较正常组降低,四君子汤高剂量组空肠内TGF-β表达较脾虚组显著升高,但与正常组亦有显著差异(P<0.05)。阳性药物和自然恢复组的TGF-β含量较脾虚组无显著差异,显著降低于正常组(P<0.05)。模型组大鼠的空肠中TNF-α的表达较正常组显著升高(P<0.05),而四君子汤低剂量组、高剂量组、阿托品组及自然恢复组的TNF-α表达均较脾虚组显著降低(P<0.05)。结论:四君子汤可以通过增加空肠内TGF-β表达,降低TNF-α表达,对利血平脾虚大鼠黏膜免疫功能有一定的调节作用,对PP结数量及其中的T淋巴细胞亚群无显著影响。
Objective: To observe the effects of Sijunzi Formula on jejunum mucosal immune function of spleen deficiency rats induced by reserpine. Method: Spleen deficiency rats, treated with Sijunzi Formula in two level of dose separately, were injected with reserpine intraperitoneally, and Atropine and blank groups are used. Peyer's patch is counted, subtype of T lymphocyte in PP is detected with flow cytometer, expression of TGF-β and TNF-α in jejunum is detected with method of imunohistochemistry staining. Results: There is no significant difference between all groups in Peyer' s patch number and subtype of T cells in PP. Sijunzi formula reversed the increase of TNF-aand decrease of TGF-β in jejunum of spleen deficiency rats to normal level significantly. Conclusion: Sijunzi formula can increase expression of TGF-β and reduce TNF - α in jejunum in different degree.
