摘要
目的探讨阻断协同刺激分子CD86对自然流产模型孕鼠母胎界面Caspase-3和Laminin B的表达及妊娠结局的影响。方法实验组于妊娠第4.5天腹腔注射大鼠抗小鼠CD86单克隆抗体,对照组注射大鼠同型IgG2b,而正常妊娠组不作任何处理。于妊娠第13.5天计算胚胎吸收率,用免疫组化法测定Caspase-3和Laminin B的表达。结果实验组的胚胎吸收率显著低于对照组(x2=7.441,P<0.05);实验组中Caspase-3蛋白阳性表达率明显低于对照组(P<0.05),实验组中Laminin B蛋白阳性表达率明显高于对照组(P<0.05)。结论妊娠早期阻断协同刺激分子CD86可使母胎界面中的Caspase-3和Laminin B分别通过各自不同的途径来发挥免疫耐受作用,并使自然流产模型孕鼠的胚胎吸收率降低至正常妊娠水平。
Objective: To investigate the effect of blockade of co - stimulatory signal CD86 on the expression of Caspase -3 and Laminin B proteins at the maternal -fetal interface and pregnancy outcome in abortion- prone routine model.Methods : Female pregnant CBA/J mice of experimental group and control group were received an i. p injection of 100μg of anti -mouse CD86 mAb in 200 μl PBS and irrelevant - isotype matched rat IgG2b, respectively, on the day 4.5 of gestation, however, no treatment was done for the normal pregnancy group. Pregnant CBA/J mice were killed on the day 13.5 of gestation: The embryo resorption rate was calculated, and immunohistochemical method was employed to detect the expression of Caspase - 3 and Laminin B proteins. Results : ( 1 ) The embryo resorption rate in experimental group was significantly reduced in comparison with that in control group ( x^2 = 7.441, P 〈 0. 05 ). (2) The positive expression rate of Caspase - 3 in experimental group was significantly reduced in comparison with that in control group ( P 〈 0. 05). The positive expression rate of Lami- nin B in experimental group was significantly increased in comparison with that in control group ( P 〈 0. 05 ). Conclusion: Blockade of co - stimulatory signal CD86 at the early stage of gestation can induce Caspase - 3 and Laminin B to exert immunologic tolerance by their different mechanisms and reduce the embryo resorption rate to normal pregnant level in abortion - prone murine model.
