β-胡萝卜素对巨噬细胞细胞毒作用及淋巴因子分泌的影响 被引量:1

ENHANCEMENT OF β-CAROTENE ON CYTOSTATIC ACTIVITY AND PRODUCTION OF TNF,IL-1 OF MACROPHAGE

摘要 研究了β-胡萝卜素对体外巨噬细胞(Mφ)抗肿瘤功能的调节作用。结果表明,巨噬细胞的细胞毒作用与其活化程度、靶细胞敏感性、效/靶比值(E/T)有关;10^(-6)mol/Lβ-胡萝卜素对巯基乙醇酸钠培养基诱导的巨噬细胞(T.G-Mφ)细胞毒阻滞效应无显著作用,但显著提高脂多糖(LPS)活化的T.G-Mφ的细胞毒作用,而10^(-6)mol/Lβ-胡萝卜素与LPS联合应用则可显著增加TNF、IL-1的产生。提示β-胡萝卜素可通过增加Mφ淋巴因子的产生,促进Mφ的抗肿瘤活性。 Effects of β-carotene on cytostasis of peritoneal macrophages were studied. Normal peritoneal Mφ,thioglycollate broth induced Mφ(T.G-Mφ)and LPS-activated T.G-Mφ were incubated with different tumor cell lines for 24h for prelinlilary cytostasis assay.The results showed that cytostatic activity of Mφ was related with its statue of activity、sensitivity of target cell and ratio of effect cell and target cell(E/T).Normal Mφ enhanced EAC cell proliferation, while T. G-Mφ inhibited EAC cell proliferation at certain E/T (beyond 25: 1),LPS activated Mφ had higher cytostatic activity than T.G-induced Mφ even at low E/T(6.25:1), and EAC cell line was more sensitive to cytostasis of Mφ than L929 cell line.β-carotene(10-6mol/L)increased the cytostatic activity of LPS-activated T.G-Mφ,but had no effects on T.G-Mφ after 24h treatment. In addition,β-carotene(10-6mol/L)could be found enhancing the production of TNF、IL-1 of LPS-activated T.G-Mφ after 24h culture, These results elucidated that β-carotene might increase the cytostatic activity of actived Mφthrough enhancement of cytokines production.
