摘要 目的:探讨CpG ODN壳聚糖纳米粒联合重组乙肝疫苗对免疫抑制小鼠的免疫增强作用。方法:选用环磷酰胺建立免疫抑制模型小鼠。将乙肝疫苗单独或和CpG ODN或CpG ODN纳米粒经后腿胫骨前肌注射到小鼠体内,ELISA方法检测抗HBsAg IsG抗体、IL-12水平;流式细胞仪检测外周血CD4^+、CD8^+T淋巴细胞亚群。结果:CpG ODN纳米粒联合疫苗组的CD4^+细胞百分数,IL-12水平及抗HBsAg IgG抗体的含量显著高于CpG ODN联合疫苗组(P<0.05)。结论:包裹在壳聚糖纳米粒中的CpG ODN较相同剂量的CpG ODN更能增强免疫抑制小鼠对乙肝疫苗的免疫应答。 Objective: To investigate the immune responses of synthetic CpG ODN by preparing chitosan nanoparticles in immunodepressed mice to recombinant hepatitis B vaccine. Method: The model of immunity suppression mice was established by CTX. The same doses of hepatitis B vaccine with or without CpG ODN or CpG ODN nanoparticles were injected into the left tibialis anterior muscle of all the immune mice. The serum anti-HBsAg IgG and IL-12 levels in mice were measured by ELISA assays. Changes of peripheral blood T lymphocyte subsets CD4^+ , CD8^+ were detected by flow cytometry. Result: Percentages of CD4^+ , IL-12 levels, and anti- HBsAg IgG level of mice immunized with hepatitis B vaccine plus CpG ODN nanoparticles increased more than the mice injected by hepatitis B vaccine plus CpG ODN. Conclusion: These studies emphasize that Chitosan-CpG ODN delivery in biodegradable nanoparticles combining with hepatitis B vaccine can facilitate the immune response in immunodepressed mice at same doses of CpG ODN.
