人生殖道疣和子宫颈癌中病毒基因检测及超微结构研究 被引量:1

Molecular and Ultrastructural Studies on Biopsies from the Patients with Cervical Carcinoma and Genital Warts

摘要 本实验用透射电镜观察了32例子宫颈癌,6例宫颈湿疣和2例外阴疣的超微结构,同时以~(32)P-dCTP标记的HPV 16 DNA为探针,检测了宫颈癌活检组织中的病毒DNA相关序列。电镜下,生殖道疣各例均可见典型凹空细胞病变,细胞核内的核小体,染色质之间颗粒,染色质周围颗粒显著增多。核酸杂交显示23例宫颈癌组织中存在病毒DNA相关序列。杂交呈强阳性及中强阳性组织中的癌细胞核中的核小体明显多于弱阳性及阴性标本。。 Thirty two biopsies from the patients with invasive squamous epithelial carcinoma of uterine cervix(INCA)from human genital warts, including 6 from cervix and 2 from vulvar, were examined by transmission electron microscopy to study the ultrastructural characteristic features of the tissues infected by HPV. HPV 16 labelled in vitro with 32P-dCTP, were used as probes to search for viral homologous sequences in biopsies from INCA. Using dot blot hybri-dization, tweenty three out of 32 INCA were positive for HPV 16 probe. The cytopathologic changes affect all layers of the epithlium on genital warts. The koilocytos can be observed in each biopsies which were characterized by a large translucent zone completely or partly surrourding the nucleus which were hyperchromatin and irregular in shape. The koilocytosis first appeared in cells of the parabasallayer, evidenfly on intermediate stratum and extended to the surrace of the epithelium. The cells of the basal and parabasal stratum were obviously hyperplasia and numerous perichromatin granules. interchromatin granules and nuclear bodies were shown in nuclei which have not been stressed sufficienty on previous reports. It appe-ared also abundant in the samples from INCA which contained HPV 16 DNA confirmed by DNA DNA hybridization, and the unclear bodies are especially more easily to be found in the specimens which are strongly positive for HPV DNA by nucleic hybridization than the weekly positive. It is suggested that nuclear bodies which were numerously distributed in nuclei are related to the infection by HPV in lower genital tracts.
