薄层色谱法检测果糖二磷酸钠和游离磷酸盐的研究

Free Phosphate and Fructose-1, 6-Diphosphoric Acid Trisodium Salt Identified by Thin Layer Chromatography

用薄层色谱法检测果糖二磷酸钠和游离磷酸盐，方法直观可靠，结果满意。采用微晶纤维素板，展开剂：正丁醇－丙酮－冰醋酸－１０％氨水－水（３５∶１５∶２０∶７．５∶２２．５），显色剂为钼酸铵混合显色液。进行果糖二磷酸钠和游离磷酸盐检测，两者的Ｒｆ值分别为０．１４及０．３５，最低检出量分别约为１２．８μｇ（Ｃ６Ｈ（１１）Ｎａ３Ｏ（１２）Ｐ２）、０．８μｇ（ＰＯ４（３－））。

A thin layer chromatographic method was used to analyze free phosphate and fructose-1, 6diphosphoric acid trisodium salt. The R_f values were 0. 35 and 0. 14 for free phosphate and fructose-1, 6-diphosphoric acid trisodium salt, respectively, on microcrystalline cellulose plate developed by a mixture of n-butyl alcohol-acetone-glacialacetic acid-10% ammonia-water(35 :15 : 20 : 7. 5 : 22. 5) . Colour developing agent was ammonium molybdate. The detection limit was 0. 8μg PO for free phosphate and 12. 8μg C_6H_11Na_3O_12P_2 for fructose-1, 6-diphosphoric acid trisodium salt.
