摘要
以抗性不同的两种小麦89122和9614为实验材料,研究了NaCl处理对小麦叶片蛋白质质量分数及结构的影响.与对照比,150 mmol/L和300 mmol/L的NaCl处理抑制89122和9614小麦种子的萌发和幼芽的生长,此效应具有浓度依赖性,但相同浓度的处理对89122产生的抑制效应明显弱于对9614小麦的抑制效应;NaCl处理使89122小麦幼苗叶片可溶性蛋白质量分数增加,却使9614小麦幼苗叶片可溶性蛋白质量分数减少;此外,傅立叶红外光谱分析结果表明,NaCl处理使两种小麦幼苗叶片蛋白的C=O键和C-N键都受不同程度的扰动.
The changes of protein content and protein structure in response to NaC1 stress were investigated in two wheat cultivars differing in salt tolerance, cv 89122 and cv 9614. The results indicated that 150 mmol/L and 300 mmol/L NaCI treatment caused a more pronounced inhibition in the sprout growth and seed germination ratio of cv 9614 compared with that of cv 89122 in a durative-dependent manner. Measurement of soluble protein content by G-250 staining showed, compared with the control, there was a significant increase of the amount of soluble protein in leaves of cv 89122 seedlings after NaCl treatment, but a decrease in cv 9614 seedlings; Coomassie Bright Blue-stained acrylamide gels further proved these results. In addition, an analysis of Fourier Transform infrared spectroscopy suggested that the change of protein structure was induced in two wheat leaves exposed to salt stress.
