摘要
介绍了一种简便快速提取香菇基因组DNA的方法,该法是对提取真菌DNA的SDS和CTAB法进行改进而成,经过修改后的SDS-CTAB法可在较短时间内高效地提取香菇基因组总DNA.制备物经琼脂糖凝胶电泳检测到大于20kb的DNA主带,基本无DNA碎带;OD260/280值显示产物纯度高,完全符合AFLP分析的要求。
This paper gives an easy and rapid preparation of genomic DNA available for AFLP analysis from Lentinula edodes, it comes with combination and some improvements from the ways of SDS and CTAB used in extracting DNA and can extract total DNA very well in short times. The DNA obtained by this method has a main DNA band more than 20kb in molecular length, which can be observed through agarose gel electrophoresis. A little DNA fragments can be seen, They have a high purity seen by their OD260/280 value and accords with the demands for AFLP analysis.
