摘要
用乳液聚合法制备了0.2μm的小颗粒聚苯乙烯及羧化聚苯乙烯胶乳,并探索了致敏的免疫胶乳在临床检验中的应用。在凝集试验方面,以弓形虫抗体检测为模式,用弓形虫抗原化学交联致敏羧化聚苯乙烯胶乳,建立了微量板法胶乳凝集试验。初步临床检测,敏感性和特异性分别为94.4%和93.3%。在免疫浊度方面,以C-反应蛋白(CRD)测定为模式,用羊抗CRP物理吸附致敏聚苯乙烯胶乳,建立了胶乳增强的免疫透射浊度测定法,在80~400μg/L范围内有良好的线性关系。
Small particles of 0.2μm diameter of polystyrene and carboxy-polystyrene latex were prepared by emulsification methods.The immuno-latex prepared by sensitization with antigen and antibody were investigated for the application in clinical laboratory.In respect of latex agglutination test,toxoplasma gondii was used as a model.T. gondii antigen was covalently bonded to carboxy-pelystyrene latex by the carbodiimide method and a latex agglutination test for antibodies to T. gondii was developed.In a preliminary clinical trial,the sensitivity and specificity were 94.4%and 93.3%respectively. In respect of turbidimetric immunoassay,C-reactive protein (CRP)was used as a model.A sheep antibody to CRP was physically absorbed to polystyrene latex,and a latex enhanced turbidimetric immunoassay was established. This assay had good linear relationship in the range of 80 to 400 μg/L.
