摘要
目的为了研究γ-生育三烯酚(γ-tocotrienol,γ-T3)对人胃癌细胞SGC-7901细胞的增殖抑制作用。方法采用细胞生长曲线,细胞核分裂,集落形成,DNA合成,Western blotting等方法,观察了不同剂量γ-T3(0,15,30和60umol/L)对SGC-7901细胞增殖及其相关细胞凋亡蛋白表达的影响。结果γ-T3可明显抑制SGC-7901细胞增殖;细胞核分裂、集落形成、DNA合成的抑制率24h分别为13.54%.77.02%、73.30%.98.36%和16.16%.65.33%;48h分别为14.15%-89.14%、90.7%-98.92%和35.1%-81.89%;Western blotting方法研究果表明,γ-T3可抑制Bcl-2蛋白表达,促进Bax蛋白表达,并诱导caspase-3蛋白产生17KD的活化片断。结论γ-T3对SGC-7901细胞生长有明显抑制作用,其抑制机制与参与调节Bcl家族相关蛋白表达有关。
Objective To investigate the effect of apoptosis on human gastric carcinoma SGC-7901 cells induced by γ- tocotrienol.Methods SGC-7901 cells were treated with different concentrations of γ-tocotrienol( 15,30 and 60 umol/L), with a negative control. The curve of cell growth, cellular mitosis, clone formatting, DNA synthesis and western blotting for correlated protein expression of apoptosis were employed. Results γ-Tocotrienol inhibited the cell proliferation. The inhibitory frequencies (IF, % ) of cellular mitosis, clone formatting and DNA synthesis were 13.54% - 77.02%, 73.30% - 98.36% and 16.16% - 65.33% at 24 h, 14.15% - 89.14% ,90.7% - 98.92% and 35.1% - 81.89% at 48 h, respectively. The further study showed γ-Tocotrienol inhibited the expression of Bcl-2 and promoted the expression of Bax, and induced the cleaved caspase-3 formation. Conclusion γ-Tocotrienol could inhibit SC, C-7901 cells prohferation. Its inhibitory mechanism may participate in adjusting the associated protein expressions of Bcls family.
