摘要
目的通过对垂体瘤转化基因(PTTG)、碱性成纤维细胞生长因子(bFGF)和基质金属蛋白酶-9(MMP-9)mRNA和蛋白在垂体腺瘤表达水平的检测和相关性分析,探讨PrTG、bFGF和MMP-9的表达与垂体腺瘤侵袭性的关系。方法30例垂体腺瘤手术标本分为侵袭组和非侵袭组,逆转录聚合酶链反应(RT-PCR)检测PTTG、bFGF和MMP-9mRNA的表达,免疫组化检测PTTG、bFGF和MMP-9蛋白的表达,分析侵袭组和非侵袭组PTTG、bFGF和MMP-9mRNA和蛋白的表达差异及三者之间的相关性。结果侵袭组垂体腺瘤PTTG、bFGF和MMP-9mRNA和蛋白表达均较非侵袭组增高,差异有显著性意义(P〈0.01)。相关性分析显示,PTTG、bFGF与MMP-9mRNA和蛋白表达相互之间均呈正相关(P〈0.01)。结论结合本实验结果及文献资料,我们认为PTTG→bFGF→MMP-9→肿瘤细胞外基质屏障突破这一调节通路可能是垂体腺瘤侵袭性产生的重要机制之一。
Objective To study the mRNA and protein expressions of pituitary tumor transforming gene (PTTG), basic fibroblast growth factor (bFGF) and matrix metalloproteinase 9 (MMP-9) in pituitary adenomas and to explore the association between the expressions and the invasiveness of pituitary adenomas. Methods The mRNA and protein expressions of PTTG, bFGF and MMP-9 were respectively determined by reverse transcription PCR (RT-PCR) and immunohistochemistry in 30 patients with pituitary adenomas who were divided into invasive and non-invasive groups. The difference in the expressions between the 2 groups and the correlation among the expressions were analyzed. Results The mRNA and protein expressions of PTTG, bFGF and MMP-9 were significantly higher in invasive group than those in non-invasive group (P〈0.01). The positive relation was found among the mRNA and protein expressions of PTTG, bFGF and MMP-9 (P〈 0.01). Conclusion The PTTG→bFGF→MM -9 pathway may play an important role in breaking down extracellular matrix and increasing the invasiveness of pituitary adenomas.
