摘要
目的对6例阿尔茨海默病(AD)患者血清二向电泳前预处理,去除高丰度蛋白,增加低丰度蛋白的检测效果。方法采用去除白蛋白球蛋白试剂盒对受试者血清进行预处理.然后二向电泳,结合质谱仪分析来鉴定兴趣蛋白质。结果通过对比AD组和对照组二向电泳图.得到24个差异蛋白质点,对这些差异蛋白点进行质谱仪分析,发现6例AD病人血清触珠蛋白表达增高,而只有3例AD病人血清转甲状腺素蛋白㈣表达降低。结论对AD血清二向电泳前预处理可以提高低丰度蛋白的检测效果,提示该方法可很好地用于鉴别AD血清生物标记。
Objective To pretreat the patients with Alzheimer's disease (AD) prior to two-dimensional gel electrophoresis (2-DE) by removing the high-abundance proteins from serum samples so as to enhance the detection of low-abundance proteins. Methods A pretrcatment with ProteoPrep Blue Albumin Depletion Kit was performed on the sera ofAD (n=6) and control (n=6 healthy elders) groups, and then 2-DE coupled to mass spectrometry (MS) was employed to identify the proteins of interest. Results After the 2-D electropherograms of AD patients and controls were compared, 24 differential protein spots were found and successfully identified as haptoglobin and transthyretin. The expression of haptoglobin was elevated in the sera of 6 AD cases, and the expression of transthyretin was decreased only in the sera of 3 AD cases. Conclusion The pretrcatment of AD serum prior to 2-DE can improve the detection effect of low-abundance proteins, indicating that it would benefit the identification ofbiomarkers in AD serum.
