摘要
为分析胸腺细胞对胸腺基质细胞功能的调节作用,将胸腺细胞与小鼠胸腺上皮细胞系(MTEC1)共育,分析胸腺细胞对MTEC1分泌IL-6的影响。结果表明,胸腺细胞能明显促进MTEC1分泌IL-6,其促进程度与两种细胞的数量及共育时间有关。胸腺细胞(4×10^6/孔)与MTEC1(1×10^5/孔)共育48小时,MTEC1分泌IL-6达高峰。去除胸腺细胞后96小时。
To analyse the influence of thymocytes on the function of thymic stromal cells , we examined the effect of fresh thymocytes on IL- 6 production by MTEC1 in coculture of hfTEC1 with thymocytes in vitro. This study provided evidences that thymocytes could significatntly increase IL-6 production by MTEC1 and the effect was in a cell dose and time of cocultured dependent fashion. The maximal effect was observed at 48hr after cocultured thymocytes(4×106/well) with MTE(1×105/well) . After thymocytes were removed from the experimental system , the thymocyte-activated MTEC1 , cultured alone for 96hr , produced 1.7-flod more IL- 6 than MTEC1 without previous interaction with thymocytes. Mitomycin-C- treated thymocytes could still promote IL-6 production by MTEC1 similarly to non-treated cells. Thyrnocytes culture supernatant and lysate had no such effect on IL-6 production by MTEC1. It suggests that thymocytes can promote MTEC1 to produce IL-6 by a mechanism in which direct interaction between thytmocytes and thymic epithelial cells is essential.
