摘要
目的:探讨核酸扩增(PCR)微板杂交捕获检测宫颈脱落细胞内高危型人乳头状病毒(HPV)感染的临床意义,建立快速、灵敏的检测方法。方法:采用PCR微板杂交捕获检测140例宫颈脱落细胞高危型HPV(HPV16/18/31/33/35/39/45/51/52/56/58/59/68)感染状况。结果:细胞学诊断低度鳞状上皮内病变(LSIL)56例,高危型HPVDNA阳性18例(阳性率为32.5%);细胞学诊断高度鳞状上皮内病变(HSIL)42例,高危型HPVDNA阳性25例(阳性率为59.5%);未明确诊断意义的不典型鳞状上皮细胞(ASCUS)31例,高危型HPVDNA阳性11例(阳性率为35.5%);鳞状细胞癌9例,高危型HPV阳性8例(阳性率88.9%)。结论:PCR微板杂交捕获检测宫颈脱落细胞内高危型HPV感染,具有快速、简便、灵敏度高、特异性强等优点,适用于大规模筛查和临床常规工作。
Objective To establish a rapid and sensitive method of detect cervical hisk-risk human papillomavirus infections in cervical cytological speciments. Methods HPV type 16/31/33/35/39/45/51/52/56/58/59/68 DNA were detected in 140 cervical specimens by Micro-plate Hybrid Capture Detection PCR Assys. Results In cervical cytological speciments of 56 LSIL , 18 cases were positive high-risk HPV DNA (32.5%) ;25 cases were positive high-risk HPV DNA in 42 HSIL cytological speciments (59.5 % ) ; Among 31 ASCUS specimens, 11 cases were positive high-risk HPV DNA ( 35.5 % ) ; for the other 9 cervical squmaous cell carcinomas specimens, 8 cases were positive high-risk HPV DNA(88.9% ). Conclusion Micro-plate Hybrid Capture Detection PCR Assys is a rapid, easy, sensitive and special assay to detect high-risk HPV infection with cervical cytological specimens, it can be applied for large-scale screening and clinical diagnosis.
