摘要
用乙酰胆碱受体(AChR)加完全弗氏佐剂(CFA)免疫大鼠前,鼻腔给予AChR可有效地预防实验性自身免疫性重症肌无力(EAMG)。本文结合表明,与非耐受对照组比,鼻腔AChR耐受组大鼠免疫后,Guo窝、腹股沟淋巴结(PILN)的单个核细胞(MNC)AChR特异的淋巴细胞增殖反应受到明显抑制,鼻腔耐受组PILN中AChR特异的CD4^+/CD8^-克隆增生也明显受抑制,差异均有显著性。此外。
Nasal administration of acetylcholine receptor(AChR) to experimental animals prior to immunization with AChR plus complete Freund's adjuvant(CFA) could effectively prevent clinical signs of experimental autoimmune myasthenia gravis (EAMG).We demonstrated that the suppression of AChR-specific lymphocyte proliferation responses from mononuclear cells (MNC) of popliteal and inguinal lymphnodes(PILN) in nasal AChR tolerized rats were more obvious than that in non-tolerized EAMG controlrats after immunization; the suppression of AChR -specific CD4+/CD8- clonal proliferation from PILN of nasally tolerized rats were obvious too, the statistically significant differences were found as compared to the non-tolerized control rats; the AChR- specific skin delayed-type hypersensitivity(DTH) responses were markedly decreased in nasally tolerized rats. The results suggest that the downregulation of specific T lymphocyte responses induced by nasal administration of AChR might be the basis of clinical myasthenia suppression.
