摘要
细胞凋亡(Apoptosis)发生在正常生理状态下的分化发育过程中,为胸腺内阴性选择的机制之一。本文报道分别将小鼠胸腺上皮细胞系MTEC1及其培养上清与裸鼠骨髓细胞进行共育,以染色体DNA发生规律性断裂为指标,对共育后的裸鼠骨髓细胞的凋亡状况进行分析,结果显示与MTEC1细胞共育后8小时,裸鼠骨髓细胞已发生凋亡,经细胞表型分析,凋亡的细胞可能为粒细胞和B细胞;而于DMEM中培养8小时以及与MTEC1上清共育8小时的裸鼠骨髓细胞末发生凋亡。结果表明MTEC1可主要通过细胞。细胞间相互作用诱导裸鼠骨髓细胞发生细胞凋亡,凋亡细胞可能主要为粒细胞和B细胞。
Apoptosis takes place under normal physiological condition. lt is most often found during normal tissue turnover, embryogenesis, metamorphosis , and endocrine dependent tissue atrophy. The inference has been made that cell death under these condition is programmed that it is a normal kinetic function of the cell, tissue ,or organ. Apoptosis is one of mechanism in the thymic negative selection. Here we reported that the mouse thymic epithelial cell line MTEC 1 that induced differentiation of nude mouse bone marrow cells in vitro could also mediate nude mouse bone marrow cell apoptosis during 8 hours of cocultivation. Thymocytes could be spontaneously fragmented (no treatment ) and occured during 8 hours of culture in vitro.
