自发性高血压大鼠室周灰质中NOS阳性神经元的形态学观察 被引量:2

摘要 【目的】探讨自发性高血压大鼠(SHR)脑室与中脑水管周围灰质中一氧化氮合酶(NOS)阳性神经元的变化及其与高血压病的可能关系。【方法】选10月龄成年SHR大鼠12只,同龄正常血压对照大鼠(Wistar-Kyoto,WKY)12只,灌注固定后取脑和脑干作冠状位冰冻切片,行NADPH-d组织化学方法染色,光镜观察和图象分析。【结果】(1)NOS阳性神经元主要分布在中脑水管周围灰质(PAG)的背外侧部,下丘脑第三脑室(3V)室周灰质,以及第四脑室(4V)室底灰质中;在3V周围,NOS阳性神经元散在分布,并可区分为室管膜下(远位)、膜内和膜上3种;左、右侧脑室周围未见阳性神经元分布。(2)SHR组上述部位的NOS阳性神经元数量较少,且平均灰度值增加(染色变浅),两组间比较具有统计学意义(P<0.05)。【结论】SHR室周灰质中NOS阳性神经元数量减少、NOS水平下降,NO合成减少,可能参与了高血压病的发病过程。 [Objective] To investigate the morphological changes of nitric oxide synthase (NOS) positive neurons in periventricular grey and midbrain periaqueductal grey of SHR, as well as the relationship with hypertension. [Method] The adult SHR (10 month old, n = 12) and WKY (10 month old, n = 12) were affnsed with 4% polyformadehyde. The coronal slices of all brains and stems were prepared and stained with NADPH-d histochemical method. The positive neurons were observed and analyzed by light microscope and image analyzer. [Results] (1) The NOS positive neurons were mainly located at the dorsolateral region of the midbrain periaqueductal grey, the periventricular grey of hypothalamns third ventricle (3V), and the subventricular grey of fourth ventricle. In the peri-3V, the positive neurons scattered and could he distinguished as 3 sorts of sub- , inner- , and epi - ependyma. No positive neuron was seen around the 2 lateral ventricles. (2) The number of NOS positive neurons of above 3 regions of SHR descended and the grey (light) degree of the ~ increased. The difference had statistically significance ( P 〈0.05). [Conclusion] The NOS positive neurons in periventricular greys of SHR decrease and NOS level descend, which may take part in the pathogenesis of the hypertension.
