补阳还五汤及其拆方对大鼠脑缺血后脑组织IL-6含量的影响

The Effect of Buyanghuawa Decoction and the Separate Compronents on the Level of IL-6 in Brain Tissue in Rats after Cerebral Ischemia

目的探讨补阳还五汤及其拆方对大鼠脑缺血后脑组织IL-6含量的影响。方法采用线栓法制备大鼠局灶性脑缺血模型。将SD大鼠随机分为假手术组、模型组、补阳还五汤组、黄芪组、活血组,采用ELISA法检测各组大鼠脑组织IL-6含量的变化。结果假手术组脑组织中未检测出IL-6;与假手术组相比,模型组脑组织IL-6含量升高明显;补阳还五汤组、黄芪组、活血组缺血侧脑组织IL-6含量较模型组升高,与拆方组比较,总方组的作用最强。结论补阳还五汤及其拆方可增加抗炎症细胞因子IL-6的含量,从而减轻缺血后的炎症反应,这可能是补阳还五汤对脑缺血损伤起保护作用的机理之一。总方组的作用明显优于其拆方组,说明方中益气、活血中药配伍具有协同作用。

Objective：To investigate the effects of Buyanghuanwu Decoction（BD, 补阳还五汤） and the separate compronents on the content of IL - 6 in brain tissue in rats after cerebral ischemia. Methods： The string inserting method according to Longa＇ s report was employed to make the model of focal cerebral ischemia. SD rats were divided randomly into the sham- operated group, the model group, the BD group, the astragus group and the blood- activating group. The content of IL- 6 were measured by enzyme- linked immunosorbent assay （ELISA） methods. Results： There was no detectable level of IL - 6 in brain tissue of the sham - operated group, Compared with that in the sham - operated group, the content of IL - 6 in model group increased significantly. Compared with that in model group, The content of IL- 6 in the BD group, the astragus group and the blood - activating group were all higher than that in the model group, the BD group had the strongest effects compared with the astragus group and the blood - activating group. Conclusion： BD could increase the content of IL - 6, then alleviate the inflammary reaction after cerebral ischemia, which may be one of mechanisms of protective effects of Buyanghuanwu Decoction on cerebral ischemia injury. Effect of BD was better than that of separate components groups, which showed that combined use of blood - activating drug and qi reinforcing drug had coordinate effect.
