摘要
通过正交实验研究黑藻中多糖的提取方法,并对其纯化鉴定.用热水提取黑藻多糖,经乙醇沉淀,活性碳脱色,Sevage法去蛋白,DEAE-Cellulose离子交换和Sephadex G-25柱层析纯化,电泳鉴定纯度和纸层析分析其组成.最佳提取条件为80℃,20倍体积的水,提取3 h,所得多糖电泳为单一组分,层析证明由D-葡萄糖和D-半乳糖构成.
To study the isolation technology, purification and compositon of polysaccharide from H. verticillata by orthogonal experiment. The polysaccharide was extracted with hot water, depigmentation with acticarbon, precipitated with ethanol. The best extraction condition was ascertained by perpendicular experiments. The protein in polysaccharide was removed by Sevage's way. The polysaccharide was Purified by DEAE- Cellulose and Sephadex G- 25 chromatogram, The purifies of which were identified by the cellulose acetate membrane electrophoresis, its compopents were analysed by paper chromatography. The best isolating condition was at 80℃ ,in 20 times water and for 3 hours extraction. It was proved that the polysaccharide was homogeneous and was composed of D- glucose and D - galactose.
