早期短期给予caspase抑制剂对于急性心肌梗死大鼠心功能影响的实验研究

摘要 目的评价早期、短期给予caspase抑制剂对于急性心肌梗死大鼠心功能的影响,并探讨其机制。方法将Wistar大鼠随机分为3组心肌梗死组、治疗组、假手术组。应用超声心动图评价心功能;caspase-3活性检测试剂盒测定caspase-3活性;TUNEL法检测心肌细胞凋亡情况。结果治疗组较心肌梗死组左室舒张末期直径(LVEDd)、左室收缩末期直径(LVESd)明显降低,而射血分数(EF)、左室短轴缩短率(FS)明显升高。治疗组较心肌梗死组caspase-3活性及凋亡指数(AI)明显降低。结论早期、短期给予caspase抑制剂可以有效改善心肌梗死大鼠心功能。抑制心肌细胞凋亡和改善心室重构是其主要作用机制。 Objective To evaluate the effect of short - term caspase inhibitor treatment as soon as possible on cardiac function and in acutemyocardial infarction rats and to probe into its mechanism. Methods Wistar rats were randomly divided into 3 groups: myocardial infarction group, treatment group, sham - operated group. The cardiac function was measured by echocardiogram. The caspase - 3 activity was detected by caspase- 3 activity assay kit. Cardiomyocyte apoptosis was detected by TUNEL. Results The left ventricular diastolic dimension ( LVEDd) and the left ventricular end systolic dimension ( LVESd) were significantly lower, while fraction shortening ( FS) and ejection fraction ( EF) were markedly higher in treatment group when compared with myocardial infarction group. The caspase - 3 activity and apoptosis index ( AI) in treatment group were reduced significantly in comparison with that in myocardial infarction group. Conclusion Short -term caspase inhibitor treatment as soon as possible can effectively improve the cardiac function in acute myocardial infarction rats and this may contribute to the inhibition of apoptosis and the amelioration of remodeling in heart.
