摘要
以鸡新城疫病毒La Sota株、传染性支气管炎病毒H120株和减蛋综合征病毒EDS-76株作为制备疫苗的抗原,以吐温80、油酸和丙二醇作为外部水相乳化剂,采用正交试验法,筛选了外部水相乳化剂的理想配比,并制备鸡新城疫-传染性支气管炎-减蛋综合征三联复乳疫苗,测定了疫苗的性状、黏稠度和稳定性。结果表明,疫苗的黏稠度为6 s±0.5 s,疫苗经3 000 r/min离心20 min,未出现分层现象,研制的疫苗是一种黏稠度较低、稳定性良好的乳白色W/O/W型复乳疫苗。
A new multiple emulsion inactivated vaccine was developed using newcastle virus La Sota strain, infectious bronchitis virus H120 strain and egg drop syndrome virus(EDS-76) as antigens. Tween 80, oleic acid and propylene glycol were used in external aqueous phase emulsifier; and the best match of external aqueous phase emulsifier was screened by orthogonal experiment method. The physical characterization, thickness, stability and safety of the vaccine were tested. The results showed that its viscosity was 6±0. 5 seconds, and it wasn't layered by centrifuge with 3 000 r/min for twenty minutes, the new developmental vaccine was a kind of w/o/w type multiple emulsion inactivated vaccine with lower thickness and better stability.
