摘要
目的观察两肾一夹肾血管性高血压发生发展过程中大鼠心肌TNF-α表达变化。方法实验采用两肾一夹(two kidney one clip,2K1C)的方法建立肾血管性高血压(renovascular hypertension,RVH)大鼠模型,将大鼠随机分为假手术组、2K1C实验组。采用鼠尾动脉测压法记录每周血压变化;在1、4、8、12周末取心脏,并计算心脏指数;ELISA法和免疫组织化学法,分别测定肾血管性高血压发生发展过程中各组心肌组织TNF-α的表达。结果在肾血管性高血压形成、发展过程及其趋向稳定的早期,大鼠心肌中TNF-α表达随血压升高而增加,与假手术组比较差异均有显著性;当高血压形成进入相对稳定时达最高峰,而高血压相对稳定后,TNF-α表达转而开始下降至低于早期。结论心肌TNF-α的表达与肾血管性高血压形成与发展相关。
Objective To observe expression of tumor necrosis factor - alpha (TNF - α) in the ventricle myocardium of the renovascular hypertension rats. Methods A silver clip was placed on the left renalartery of Spra- gue - Dawley rats. After the operation the rats were divided into two groups: sham group and 2K1 C( two - kidney one clip)group. Caudal vein blood pressure was measured every week..Maimals were respectively killed after one, four, eight, twelve weeks. Then cardiac index( CI)was calculated, and TNF-α protein in ventricle myocardium was measured by ELISA and immunohistochemistry. Results The amount of myocardial TNF - α in 2K1C group was significandy higher than that in sham group. TNF- α increased significantly in the early development of renovascular hypertension, and it reached the peak when the renovascular hypertension stablized, and then declined. TNF-α plays an important role during the formation and development of renovascular hypertensive.
