一种快速响应的压电晶体生物亲和传感器

摘要 在6MHz的压电晶振QCM(quartz crystal microbalance)金电极表面采用蛋白质A—金的方法固定生物敏感膜,利用QCM单面测试技术和连续流动自动分析系统全面测试分析此膜与人体免疫球蛋白G(IgG)之间的生物亲和反应,从而研究快速响应的压电晶体生物亲和传感器。停止流动法测量结果为:在1—2min内亲和响应随时间而急剧增大,随后趋于缓慢,对107μg/ml人体IgG三次响应的标准偏差及变异系数分别为2.5Hz和2.77%,在1—107μg/ml测量范围内响应值与测试浓度对数呈线性关系,用关系数和灵敏度分别为0.9824和53.6Hz/dec.(μg/ml):连续流动法测试结果为:进样时间约为1min,每个样品测试时间为3min,在2.9-249μg/ml测量范围内响应值与测试浓度对数呈线性关系,相关系数和灵敏度分别为0.9797和68.9Hz/dec.(μg/ml)。 The bioaffinity sensing film was immobilized by protein A-gold method on the gold electrod surface of 6MHz quartz crystal microbalance(QCM). The QCM bioaffinity sensor with rapid response was studied by using QCM one-side technique and the continuous-flow auto-analytic measurement system, testing and analyzing the bioaffinity sensing of protein A and human IgG. By using stop-flow method, the speed of bioaffinity was found to be the most fast within 1 - 2min at beginning; The standard deviation and variation coefficient for 3 times performances response to 107μg/ml human IgG were found to be 2. 5Hz and 2. 77% respectively, A linear.ar concentration range of 1 - 107 fig/ml on logarithmic scales was acquired, the correlation coefficient and response sensitivity were0. 9824 and 53. 6Hz dec. (μg/ml) respectively. By using continuous-flow method, the sampling time and the test time per sample were found to be 1min and 3min respectively. A linear concentration range of 2. 9 - 249μg/ml on logarithmic scales was acquired, the correlation coefficient and response sensitivity were 0. 9797 and 68. 9Hz/dec. (μg/ml)respectively.
