摘要
目的通过原发性胆汁性肝硬化患者肝组织和外周血单个核细胞Fas、FasL的表达,探讨Fas、FasL在原发性胆汁性肝硬化患者发病机制中的作用。方法应用免疫组织化学技术,以兔抗Fas及兔抗FasL多克隆抗体,对20例原发性胆汁性肝硬化(PBC)患者肝组织和外周血单个核细胞中的Fas、FasL进行检测。结果18例PBC患者外周血单个核细胞检出Fas、FasL,检出率均为90%,Fas、FasL平均阳性细胞数分别为62%、59%。20例PBC患者16例Fas阳性颗粒表达于肝细胞胞浆,4例表达于肝细胞胞膜;20例FasL棕黄色阳性颗粒均表达于肝细胞胞浆,Fas、FasL在肝组织中浸润的淋巴细胞胞浆有表达,并多聚集于汇管区周围。结论PBC患者的Fas、FasL表达增加,它们在PBC的发病机制中发挥重要作用。
Objective To study the pathogenesis role of Fas and FasL in PBC patients, we expressed their Fas and FasL in liver tissues and PBMC.Methods There were 20 PBC were entranced. Immanohistochemistry was used to detecte Fas and FasL in liver tissues and PBMC.Resuits 18 cases detected Fas and FasL in PBMC of 10 PBC,and the average percent of positive cells of Fasand FasL were 62% and 59%,respectively.With 20 PBC patients,the positive rates for Fas and FasL in liver tissue were all 100%.Fas in 16 cases were loated in cytoplasm and 4 in cell membrane. The distribution of Fas positive hepatocyte was in the enviroment of part; FasL were in the cytoplasm.FasL positive hepatocytes were also in the enviroment of port. Conclusion The expressions of Fas and FasL were increased in PBC compared with normal control, and they may play an import role in the pathogenesis of PBC.
