L-精氨酸对HeLa细胞端粒酶活性的影响

摘要 目的:探讨L-精氨酸对HeLa细胞端粒酶活性的影响。方法:采用端粒酶重复序列扩增法-银染法检测端粒酶活性的变化。结果:L-精氨酸浓度低于1mmol/L组,端粒酶活性没有明显改变;5~25mmol/L组,端粒酶活性显著增强;50~100mmol/L组,端粒酶活性显著受到抑制。不同浓度L-精氨酸室温处理HeLa细胞提取液2h后,端粒酶活性与对照组无显著差异。结论:体内L-精氨酸浓度的变化引起端粒酶活性的改变,提示L-精氨酸引起的肿瘤发生发展与端粒酶活性改变有关。 Objective:To investigate the effect of L-arginine on the telomerase activity in HeLa cells. Methods:HeLa cells were treated with different concentrations of L-arginine. Telomerase repeat amplification protocol (TRAP)-silver staining was used to detect the activity of telomerase. Results:The activity of telomerase did not change when the concentration of L-arginine was less than 1 mmol/L, increased when the concentration of L-arginine ranged from 5 to 25 mmol/L, and was inhibited when the concentration of L-arginine ranged from 50 to 100 mmol/L. No significant difference in telomerase activity was found between HeLa cells treated with different concentrations of L-arginine for 2 hours and those not treated with L-arginine. Conclusion:3he change of L-arginine concentration may alter the telomerase activity in vivo, which indicates the development of carcinoma induced by L-arginine may relate to the alteration of telomerase activity.
