利用MS2噬菌体包膜蛋白与RNA的特异性相互作用构建临床检测用病毒样颗粒 被引量:1

摘要 MS2噬菌体是单链正链RNA病毒,为26nm长的20面体,共有3569个核苷酸,包含有3个基因,编码成熟酶蛋白、包膜蛋白、复制酶蛋白和裂解蛋白等4种蛋白质分子。噬菌体由180个包膜蛋白单体、一分子成熟酶蛋白和一分子基因组RNA组成。在MS2噬菌体包膜蛋白的研究中发现包膜蛋白与噬菌体复制酶5’端由19个碱基(19mer)组成的茎环结构RNA序列有特异性相互作用,这种作用可引发噬菌体外壳的组装,同时,将噬菌体基因组RNA包装到包膜内。国外研究人员发现在包装位点后引入非噬菌体基因序列也可以引发包装。本研究拟利用MS2包膜蛋白与RNA的相互作用来观察RNA序列对包装的影响以及基于两者相互作用构建病毒样颗粒在临床分子检测中的应用。
