摘要
目的建立测定蒙药森登-4汤中栀子苷、槲皮素和没食子酸的高效液相色谱方法。方法栀子苷的测定条件为:Hypersil C18 ODS(250.0 mm×4.6 mm,5μm)色谱柱,流动相为乙腈-水(体积比为13.5∶86.5),流速为0.8 mL.min-1,检测波长为238 nm。槲皮素的测定条件为:Century SIL C18 BDS(250.0 mm×4.6 mm,5μm)色谱柱,流动相为甲醇-水-磷酸(体积比为60.00∶40.00∶0.02),流速为0.8 mL.min-1,检测波长为360 nm;没食子酸的测定条件为:Century SIL C18 BDS(250.0 mm×4.6 mm,5μm)色谱柱,流动相为乙腈-水-磷酸(体积比为5.00∶95.00∶0.02),流速为0.80 mL.min-1,检测波长为272 nm。结果在各自的色谱条件下,栀子苷、槲皮素和没食子酸质量浓度与峰面积具有良好的线性关系。平均回收质量分数分别为:栀子苷96.9%(RSD=1.9%,n=6);槲皮素97.1%(RSD=2.3%,n=6);没食子酸98.7%(RSD=1.7%,n=6)。结论该方法可作为该制剂的含量测定方法。
Objective To establish the HPLC method for determination of geniposide, quercetin and gallic acid in Mongolian medicine Sendeng-4 decoction. Method The determination of geniposide was performed with column Hypersil ODS C18(250 mm × 4.6 mm, 5 μm) using acetonitrile-water( V : V = 13.5 : 86.5) as mobile phase at a flow rate of 0.8 mL·min^- 1. UV detective wavelength was set at 238 nm. The determination of quercitin was performed on Century SIL Cls BDS (250 mm × 4.6 mm, 5 μm)column, using methanolwater-phosphoric acid ( V: V : V = 60.00 : 40.00 : 0.02) as mobile phase at a flow rate of 0.8 mL· min^- 1 and UV detection at 360 nm. The determination of gallic acid was performed on Century SIL C18 BDS (250 mm×4.6 mm, 5 μm) column, using acetonitrile-water-phosphoric acid ( V : V : V = 5000 : 95.00 : 0.02) as mobile phase at a flow rate of 0.8 mL· min^- 1 and UV detection at 272 nm. Results The standard curves of geniposide, quercetin, and gallic acid had a good linearity under the HPLC conditions. The average recovery and relative deviation were 97.1% with 2.3 % for geniposide, 96.9 % with 1.9 % for quercetin, and 98.7% with 1.7 % for gallic acid, respectively. Conclusion The methods are simple, accurate, and can be used as methods for the determination of Sendeng-4 decoction.
