摘要
目的探讨知母总皂苷(total timesaponin,TT)抗实验性抑郁作用并分析其可能的作用机制。方法以小鼠强迫游泳实验及悬尾实验为动物模型,考察TT抗实验性抑郁作用;以皮质酮诱导PC12细胞损伤为细胞模型,采用细胞形态学方法、MTT法、LDH法观察TT对PC12细胞损伤的保护作用。结果TT(25、50 mg.kg-1)能显著缩短小鼠不动时间,且对自主活动无明显影响。TT(0.05、0.50、5.00 mg.L-1)能明显改善皮质酮诱导的PC12细胞形态的改变,显著提高细胞存活率(P<0.01),并减少LDH的外漏(P<0.01)。结论TT具有一定的抗实验性抑郁作用,其机制部分可能是对抗皮质酮诱导的细胞损伤。
Objective To study the antidepressant effects of total timesaponin (TT) on experimental depression models and analyze the mechanism of this action. Methods Using forced swimming test (FST) and tail suspension test (TST) as in vivo model, the antidepressant activities of TT were investigated. Using PC12 cells damage induced by corticosterone as in vitro model, the protective effects of TT were determined through morphological examination, MTT assay and LDH assay. Results Chronic treatment of TT (25, 50 mg· kg^-1) significantly decreased the duration of immobility in the FST and TST, and locomotor activity was not affected. TT (0.05,0.50, 5.00 mg·L^- 1) could improve the morphological change induced by corticosterone, markedly increase the survival rate (P 〈 0.01) and decrease the LDH activity (P 〈 0.01). Conclusion TT has antidepressant activity in behavioral models that might be mediated via the cytoprotective action shown in PC12 cells.
