适于磁共振功能成像研究的急性癫痫猫模型的建立

摘要 目的建立适于脑功能研究的急性癫痫动物模型。方法家猫10只,随机分为两组,实验组肌注戊四氮(PTZ)55mg/kg后观察其行为表现,进行脑电图检查,并分别于癫痫发作前1h,发作后1h、1d、3d、5d、7d行MRI检查。对照组肌注等量生理盐水。结果实验组均于注射PTZ后1~5min出现肢体节律性抽动及全身阵挛,脑电图显示有特征性尖波、棘波、棘波群,MR上均无明显异常发现。对照组无异常行为及心电图变化。结论该急性模型建立快,成功率高,有良好的可预测性和可重复性,无明显脑结构性异常,是进行脑功能研究的理想模型。 Objective To establish cat model about acute epilepsy which be suited for brain function study. Methods 10 cats were arbitrarily divided into 2 groups. Cats of experimental group were injected pentylenetetrazol (PTZ)(55mg/kg) intramuscular, and those of contorl group were injected equivalence saline. Behavior change were observed, electroencephalogram were undertakn and MRI were performed. Results After injection PTZ intramuscular, epileptic seizure were all evoked, EEG recording showed spike-wave and polyspike-wave complexes. There were no significant abnormal finding on MRI. Cats of contorl group had no behavior and electroencephalogram change. Condusion The model has excellent stability and reproducibility and is suitable for experimental studies of cerebral function.
