雷公藤内酯醇对大鼠局灶性脑缺血再灌注损伤的拮抗作用 被引量:4

摘要 目的:探讨雷公藤内酯醇对大鼠局灶性缺血再灌注脑损伤的保护作用。方法:采用大鼠局灶性脑缺血再灌注模型,雷公藤内酯醇0.2、0.4mg/kgip后,检测大脑中动脉闭塞侧的梗死体积、神经功能受损程度、脑组织含水量、脑微血管内中性粒细胞计数。同时光镜下观察脑组织损伤程度和脑神经细胞IL-1β免疫组化染色改变。结果:与模型组比较,雷公藤内酯醇两治疗组大脑中动脉闭塞侧脑梗死体积、脑含水量、微血管内中性粒细胞计数及光镜下脑神经细胞受损程度均明显减少或减轻。脑神经细胞IL-1β免疫组化染色阳性细胞数明显减少,神经功能受损程度也明显改善。结论:雷公藤内酯醇对大鼠缺血再灌注脑损伤有保护作用。 Objective: To investigate the protect effect of triptolide ( TL, at dose 0.2,0.4mg/kg, ip) on cerebral of rats after focal ischemic reperfusion. Methods: The focal cerebral ischemic-reperfusion model was established with thread occlusion of middle cerebral artery. We observed the effects of embolism of middle cerebral atery. We observed the effects of TL after focal ischemic reperfusion on the volume of middle cerebral after infarction, the change of cerebral water content, leukocyte accumulation adhesion in micro-vascular and damage degree of brain function ; meanwhile the pathology change of brain tissue was observed with light micrascope. Immunohistochemical techniques was used to evaluate IL-1 β express in cortex.. Results: Compared with the model group, the volume of middle cerebral after infartion, the water content of cerebral, and accumulation of leukocyte in micro-vascular were remarkably reduced in two group of TL-treated. The pathology injury degree of cerebral tissue was significant improved under light microscopy. The expression of IL-1 β in cortex were significantly decreased. Conclusion: The results suggested that TL had a potent neuroprotective effect in cerebral ischemial and repeffusion in rats, the mechanism related to reducing volume of cerebral infarction and inhibited the infiltration of neutrophil in the cerebral tissues of rats.
