摘要
把溶于双蒸水的脱氧核糖核酸(DNA)铺展在金膜上,再覆盖一薄层50%甘油水溶液。用扫描隧道显微镜的恒定电流模式,我们成功地获得了DNA内部结构碱基对水平的高分辨图像,图中可以分辨碱基对。本工作展示了未来运用STM来研究核酸和分辨嘌呤、嘧啶的前景。
DNA dissolved in double-distilled water were deposited onto gold surface and covered with 50%glycerol-water solution.Through constant-current mode STM,we succeeded in obtaining images which we believe reveal for the first time the basepair-level structure of DNA in its near-native environment.This im-plies the possibility of identifying purines and pyrimidines in the future.
