大鼠胚胎神经干细胞的低温保存

摘要 目的建立大鼠胚胎神经干细胞(neural stem cells,NSCs)的低温保存方法,探讨经冻存后NSCs的存活及生物学特性变化。方法使用二甲基亚砜(DMSO)做NSCs的冷冻保护剂于液氮中冻存。复苏后用间接免疫荧光染色方法对细胞的存活、形态及分化能力做鉴定。结果不同冻存时间及代数的NSCs复苏后存活率无明显差异(P>0.05),且仍能在体外多次传代,并可分化为神经元、星形胶质细胞和少突胶质细胞。结论大鼠NSCs低温冻存、复苏并不影响其原有的形态、增殖及多向分化等生物学特性。
