脑欣口服液对新生大鼠缺氧缺血性脑损伤能量代谢影响的实验研究

摘要 目的探讨脑欣口服液对新生大鼠缺氧缺血性脑损伤能量代谢的影响。方法将7日龄SD大鼠50只随机分为假手术组、缺氧缺血性脑病(HIE)模型组和脑欣口服液大剂量组、中剂量组、小剂量组,每组各10只。HIE造模后,脑欣口服液大、中、小剂量组每只大鼠每次均灌胃0.75 mL(分别含生药0.625、0.5、0.375 g),每日2次,均给药10日;模型组和假手术组均灌服等量的0.9%氯化钠注射液,均给药10日。测定各组新生大鼠脑组织中钠、钾-三磷酸腺苷酶(Na、K+-ATP酶)、钙-三磷酸腺苷酶(Ca2+-ATP酶)及镁-三磷酸腺苷酶(Mg2+-ATP酶)活性。结果模型组与假手术组比较,Na、K+-ATP酶、Ca2+-ATP酶、Mg2+-ATP酶活性差异均有显著性意义(P<0.05),证明造模成功。模型组与脑欣口服液大剂量组比较,Na、K+-ATP酶、Ca2+-ATP酶、Mg2+-ATP酶活性差异均有显著性意义(P<0.05);与脑欣口服液中剂量组比较,Na、K+-ATP酶及Ca2+-ATP酶差异均有显著性意义(P<0.05);与脑欣口服液小剂量组比较,Ca2+-ATP酶差异有显著性意义(P<0.05)。结论脑欣口服液抑制新生大鼠缺氧缺血性脑细胞凋亡的机理与Na、K+-ATP酶、Ca2+-ATP酶及Mg2+-ATP酶升高有关。 objective To explore the effects of Naoxin oral liquid on energy metabolism of rat brain injury induced by ischemia and hypoxia. Methods 50 SD rats of 7 day were divided into sham operation group, HIE model group, large dose of Chinese medicine group, medium dose of Chinese medicine group, and small dose of Chinese medicine group with 10 rats in each group, Naoxin oral liquid was fed by the same volume of normal salina from the large, medium and small dose groups after model was duplicated. Na, K^+ - ATP enzyme, Ca^2+ - ATP and Mg^2 + - ATP in each group were measured. Results There were significant difference of three indexes between sham operation group and HIE model group ( P 〈 0.05) . There were signiticant difference of three indexes between sham operation group and every treatment group ( P 〈 0.05) . Conclusion The mechanism of inhibiting apoptosis of new-born rats brain cell from ischemia and hypoxia by Naoxin oral liquid is well related to the increase of Na,K^+ - ATP enzyme , Ca^2+ - ATP enzyme and Mg^2+ ATP enzyme.
