摘要
目的:探讨具有补肾益气作用的软脉煎对与家兔胸主动脉血管平滑肌细胞增生有关的血小板源生长因子-A链(PDGF-A)mRNA以及细胞超微结构的影响。方法:用贴块法培养家兔胸主动脉血管平滑肌(VSMC),用药物血清学方法将药物血清作用于细胞模型。设立正常组,模型组,软脉煎5倍、10倍、20倍成人剂量组和舒降之10倍成人剂量组。逆转录—聚合酶联反应法测定各组血小板源生长因子A链mRNA表达水平。电镜观察各组超微结构的改变。结果:软脉煎能够使家兔VSMCs PDGF-A链的mRNA表达降低并改变其超微结构。结论:肾中精气亏虚可能是动脉粥样硬化病变发生的根本原因之一,补肾益气可能是治疗动脉粥样硬化的有效原则。
Objective：such as PDGF- A mRNA and ultrastucture of VSMCs. Methods：Rabbit aortic vascular smooth muscle cells were cultured by a substrate - attachedexplant method, serum was acted on the cell model by serum pharmacologic method. Set up normal group, model group, slmavastatin group and group of different Ruanmal Jian dosage. The expression of PDGF - A mRNA were dbserved and analysed by means of reverse transcriptase - polymerase chain reaction assay; Ultrastructure of the SMCs which were underwent diffent management was investigated by transmission dectron microscope. Result： Ruanmai Jian can influence the expression of PDGF - A mRNA and ultrastucture of VSMCs. Conclusion： Asthenia of the kidney essence and qi could be one of the basic causes resulated in AS. Supplementing kidney essence and qi could be a effectual therapeutic method.
