摘要
目的:对弓形虫上海株SAG2基因进行克隆与鉴定。方法:采用PCR方法从弓形虫上海株总DNA中扩增到SAG2基因,与pGEM-T-easy vector连接,并进行DNA测序分析。结果:用DNAStar软件对扩增出的片断与GenBank上的7个虫株相应的序列进行同源性比较,不同宿主和不同区域上的弓形虫SAG2基因序列差异很小。结论:用基因重组技术获得的SAG2抗原,作为候选疫苗和诊断抗原,有着广阔的发展前景。
In oder to get and identify SAG2 gene of Toxoplasma gonddi, SAG2 gene sequence were obtained from the DNA of Toxoplasma Shanghai strain by PCR. The gene was introduced into pGEM-T easy vector and then it was sequenced. Nucleotide sequence analysis by DNA star revealed that the homology of Toxoplasma Shanghai SAG2 gene with that of other seven Toxoplasma strains from Genbank was very high.
