摘要
目的通过检测川芎对高糖诱导血管内皮细胞Bcl-2和Caspase-3基因表达的影响,探讨川芎抑制血管内皮细胞凋亡的分子机制。方法含川芎血清培养血管内皮细胞,对照组用正常血清和无血清培养基培养。收获细胞后,提取总RNA,设计特异性引物,通过实时荧光定量PCR的方法,检测川芎对血管内皮细胞Bcl-2和Caspase-3基因表达的影响。结果含川芎血清可以显著提高Bcl-2和Cas-pase-3基因的表达,与其他组相比存在显著性差异。结论川芎可能是通过调节Bcl-2和Caspase-3基因的表达,影响Caspase凋亡信号传导系统,抑制血管内皮细胞的凋亡,达到治疗的作用,这可能是川芎抑制血管内皮细胞凋亡的分子机制之一。
OBJECTIVE To investigate the molecule mechanism of Rhizoma chuanxiong (川芎) in inhibiting apoptosis of vascular endothelial cells.METHODS Vascular endothelial cells were cultured with serum containing Rhizoma chuanxiong, a Chinese herb, in the treatment group and with normal serum and without serum in the control group. After cells were harvested, we distilled overall DNA and designed special guide. The effect of Rhizoma chuanxiong on gene expression of Bcl - 2 and Caspase - 3 was detected by fluorescence ration real time PCR. RESULTS The serum containing Rhizoma chuanxiong showed enhanced gene expression of Bcl - 2 and Caspase - 3. It showed significant difference between treatment group and other groups. CONCLUSIONS Rhizoma chuanxiong probably achieved its treatment purpose by adjusting gene expression of Bel - 2 and Caspase- 3, influencing signal transduetion system of Caspase apoptosis and inhibiting apoptosis of vascular endothelial cells.
