摘要
目的研究水飞蓟宾对NASH大鼠肝纤维化进展的影响。方法30只雄性SD大鼠随机分为正常饮食对照组(n=10)、高脂饮食模型组(n=10)和水飞蓟宾治疗组(n=10),实验时间12周。生化法检测血清AST,ALT,TG,CHO。放免法检测HA,RT-PCR法检测TIMP-1、TIMP-2、MMP-2、MMP-13 mRNA的表达,紫外分光光度计测量SOD及MDA。结果与模型组比较,水飞蓟宾治疗组大鼠肝功改善,HA及SOD含量下降,TIMP-1、TIMP-2、MMP-2表达降低,MMP-13及MDA升高,肝纤维化程度减轻。结论水飞蓟宾具有一定抗纤维化作用。
Objective In order to investigate the effect of silybin on the progression of NASH. Methods Rats were randomly divided into three groups: control group, model group and therapeutic group. The therapeutic group received silybin for 12 weeks. The level of serum AST,ALT,TG,CHO were measured by biochemistry method. Serum hyaluronic acid was measured by radio-immunity method. The expression of TIMP-1 ,TIMP-2 ,MMP2, MMP13 was detected by RTPCR. MDA and SOD was detected by UV spectrophotometer. Results Compared with the model group, there was an obvious decrease of AST, ALT, hyaluronic acid and SOD, the expression of TIMP-1 ,TIMP-2, MMP2 and MDA decreased, the expression of MMP13 was higher in the group treated by silybin. There was improvement in histology and liver fibrosiss in the group treated by silybin. Conclusion silybin could inhibit the progress of rat liver fibrosis.
