摘要
目的探讨色素上皮衍生因子(PEDF)在喉癌组织血管生成中的作用及PEDF与喉癌临床病理因素的关系。方法用免疫组织化学法检测42例人体喉鳞癌标本和11例正常喉黏膜组织中PEDF和血管内皮生长因子(VEGF)的表达,标记内皮细胞黏附分子(CD31)分子显示血管内皮细胞,并计数上述喉鳞癌组织内的微血管密度;统计学分析上述分子标记物间的关系以及各分子标记物与喉鳞癌临床病理因素间的关系。结果所有的正常喉黏膜(11例)均有PEDF表达,喉鳞癌组织PEDF表达率47%(20/42),VEGF表达率57%(24/42),淋巴结转移者呈现PEDF低表达,表达率14%(6/42)(P<0.01);喉癌组织中PEDF的表达与微血管密度(MVD)(P<0.01)和VEGF(P<0.01)的表达呈负相关关系,VEGF表达与MVD呈正相关关系(P<0.01)。结论PEDF低表达和VEGF高表达可能是喉鳞癌组织内微血管生成的分子基础,是促进喉鳞癌细胞发生淋巴及血管转移的重要因素。
Objective To investigate the role of PEDF in angiogenesis and association of PEDF expression with clinicopathological factors in laryngeal carcinoma. Methods SABC immunohistochemistry method was used to detect the expression of PEDF and VEGF in 42 cases of laryngeal carcinoma and in 11 cases of laryngeal normal tissue samples. CD31 was used to endothelial cells and the MVD was calculated. The relationship between the molecular markers and the relationship between the markers and clinicopathological factors of laryngeal carcinoma were analyzed by statistical method. Results PEDF expression was showed in all normal(ll cases) laryngeal mucous menbrane. The positive rate of PEDF and VEGF were 47% (20/42)and 57%(24/42) in laryngeal carcinoma Compared with well-differentiated laryngeal cancer. The laryngeal carcinoma cells with lower PEDF expression tend to metastases by lymphatic vessels and the positive rate were 14%(6/42) (P〈0. 01). There were negative correlation between PEDF and MVD (P〈0. 01), VEGF (P〈 0.01) and positive correlation between VEGF and MVD (P〈 0. 01). Conclusion Lower PEDF and higher VEGF expression might be the molecular basis of angiogenesis in laryngeal carcinoma tissue and play an important role in lymphatic and blood metastasis of laryngeal carcinoma.
