摘要
目的:探讨急性酒精灌胃后大鼠胃黏膜损伤的程度和胃组织中TNFα和血浆PGE2、血清NO的改变。方法:利用酒精定量灌胃制作急性酒精性胃黏膜损伤动物模型,采用小格图纸法计算不同时点(0、15、30、60分钟)大鼠胃黏膜损伤面积;同时用免疫组化ABC法和显微图像分析系统测定胃组织中TNF-α定位和光密度值,采用ELISA和硝酸还原酶法测定血中PGE2和NO。结果:酒精灌胃15分钟后,胃黏膜损伤指数即显著超过对照组的5倍(40.6对8.2,P<0.01);酒精组(各时相光密度值分别为0.2047、0.2387、0.2686、0.3110)与对照组(0.1799 ̄0.1944)相比,胃黏膜TNF-α的表达均增加,差异具有显著性(P<0.05);酒精组NO水平逐渐降低(分别为54、48.8、41.6、42.9μmol/l),对照组为52.2~59.9,饮酒15~30分钟,酒精组血浆PGE2水平(10、11pg/ml)与对照组(15pg/ml)相比均有明显降低(P<0.01)。结论:短期内酗酒可造成胃黏膜的高度损伤,并诱发胃黏膜TNF-α表达增加,同时降低血中NO、PGE2水平。
Objective:To study the changes of gastric mucosa and the expression of tissue TNFct,plasma PGE2 and serum NO in rat which received ethanol orally.Methods:Making animal model of acute gastric mucosa injury by ethanol which was intragastriced in fix quantity,using chart paper to calculate the area of injured gastric mucosa at different time (0.15.30.60 min);determining the TNF-α lo-ation and optical density value in gastric mucosa by ABC immunochemistry method and microimage pattern analytical system.And determining the plasma PGE2 and serum NO by ELISA method and nitrate reducing enzyme method.Results:15min after ethanol orally,the injury index of gastric mucosa is 5 times higher than the control group (40.6 to 8.2,P〈0.01);the expression of tissue TNF-αin the ethanol greup(the optical density value of every phase is 0.2074,0.2387,0.3110)is higher than the control greup(0.1799-0.1944),possessing significant difference (P〈0.05);The level of NO decreased gradually in ethanol group (54,48.8, 41.6,42.9 μmol/l respectively),however,the control group is ranged from 52.2 to 59.9 Ixmol/L.15-30min later, the level of plasma PGE2 (10-11pg/nd)is lower apparently (P〈0.01) than the control group (15pg/ml).Condusion:Alcohol abuse in short term can cause serious injury on gastric mucosa and increase the expression of tissue TNF-α,meanwhile,decrease the level of plasma NO and serum PGF-E2.
