摘要
目的研究凋亡相关基因诱骗受体3(DcR3)在系统性红斑狼疮患者(SLE)外周血单个核细胞(PBMCs)中的mRNA表达水平。方法应用半定量逆转录-聚合酶链反应(RT-PCR)检测38例SLE患者(21例活动期,17例缓解期)和15例正常人PBMCs中DcR3 mRNA的表达水平,分析其与狼疮活动指数(SLEDAI)的相关性。结果SLE患者PBMCs中,DcR3mRNA表达水平明显高于正常人(P<0.01),但疾病不同病期(活动期与缓解期)其表达水平无统计学差异(P>0.05);DcR3 mRNA表达与SLEDAI不相关(r=-0.024,P>0.05)。结论SLE患者PBMCs DcR3 mRNA表达增加,提示DcR3可能与SLE免疫细胞凋亡异常和免疫调节的紊乱有关,参与了SLE的发病过程。
Objective To investigate the expression of DcR3 mRNA in peripheral blood nuclear cells (PBMCs) from patients with systemic lupus erythematosus (SLE). Methods The expression of DcR3 mRNA in PBMCs from 38 patients with SLE(21 active and 17 inactive)and 15 cases of heahhy subjects was measured by reverse transcription- polymerase chain reaction ( RT- PCR). The correlation was analyzed between DcR3 mRNA expression level and Systemic Lupus Erythematosus Disease Activity Index (SLEDAI). Results The DcR3 mRNA expressed in SLE was significantly higher compared with healthy controls (P 〈0.01 ),whereas no statistical difference between the active and inactive SLE patients was observed( P 〉 0.05 ). The relationship between expression of DcR3 mRNA and SLEDAI scores was not found( r = - 0. 024, P 〉 0.05 ). Conclusion The expression of DcR3 mRNA in PBMCs from patients with SLE is significantly increased, perhaps indicating that DcR3 is involved in the abnormal immune reactions and the pathogenesis of SLE.
