高效液相色谱法测定大籽獐牙菜中有效成分的含量

Determination of the Contents of Effective Constituents in Swertia macrosperma C.B. Clark by HPLC

目的:建立以高效液相色谱法测定大籽獐牙菜中1-羟基-3,7,8-三甲氧基酮、1,5,8-三羟基-3-氧基酮、1,8-二羟基-3,7-二甲氧基)酮和1,8-二羟基-3,5-二甲氧基酮含量的方法。方法:色谱柱为VP-ODSC18(150mm×4.60mm,5μm),流动相为甲醇-水-磷酸(70:30:0.4),检测波长为254nm,流速为1.0mL.min-1,柱温为20℃。结果:1-羟-基3,7,8-三甲氧基酮、1,5,8-三羟基-3-甲氧基酮、1,8-二羟基-3,7-二甲氧基酮和1,8-二羟基-3,5-二甲氧基咄酮进样量分别在0.172～0.86、0.224～1.344、0.144～0.72、0.154～0.77μg范围内线性关系良好,平均回收率(RSD)分别为98.18%(1.19%)、99.32%(1.52%)、98.44%(1.23%)、97.68%(1.57%)。结论:本方法快速、简便,结果准确、可靠。

OBJECTIVE： To establish an HPLC method for the determination of the contents of Decussatin, Bellidifodin, Methylswertianin and Swerchirin in Swertia macrosperma C. B. Clark. METHODS： The determination was performed on VP - ODS C18（150 mm ×4.60mm,5μm）, with the mobile phase consisting of methanol- water- phosphoric acid（70 ： 30 ： 0.4）, the detection wavelength set at 254 nm, the flow rate at 1.0 mL · min^ -1 and the column temperature at 20℃. RESULTS： The calibration curves of Decussatlng, Bellidifodin, Methylswertianin and Swerchirin were in good linearity in the range of 0.172-0.86 μg, 0. 224-1. 344μg, 0.144-0.72μg and 0.154- 0.77μg, respectively.The average recoveries were 98.18% （RSD= 1.19% ）, 99.32% （1.52%）, 98.44% （1.23%） and 97.68% （1.57%）, respectively. CONCLUSION： The method is rapid and precise, with accurate and reliable results.
